3. KET QUA VA THAO LUAN

Summary The aims of the study were to select the strain of Saccbaromyces cerevisiae and optimize the conditions for biomass production in tofu sour liquid medium, and to invesUgate the conditions for the acquisition of total yeast extract and p-glucan from yeast biomass. 5. cerevisiae LXl.l was selected with high biomass production (0.204 g/100 mL) in tofu sour liquid at pH 5.53 from 10 tested yeast strams. Ey experimental Plackett-Burman design (PBD), 7 factors affecting the yeast autolysis were investigated. Tliree factors including pH, initial cell level and peptone concentraUon which statistically significant affected on yeast biomass were continued lo optimize using CenUal Composite design (CCD), The results showed that the suitable growth conditions for 5 ce/'ews/ae LXl.l were determined at 10' cells/mL, 4.78 g/L supplemented peptone and pH 5.59. the dried biomass was obtained at 0.469 g/ lOOmL In total. 0.298 g of yeast extract and 2.583 g of p-glucan were achieved from 3.0 g of dried yeast cells and the recovery ratio of p-glucan was obtained at 86.12%.
