RATS.

Abstract

This study was carried out to investigate the toxicity of the oral administration of

uranyl acetate on kidney. For this purpose, twenty females mature Albino rats which

were divided into four equal groups, control group (G1) administrated distilled

water, and other three groups administrated orally for 70 days (10 weeks), with (50,

75 and 100 mg/kg/b.w. /day of uranyl acetate), for 10 days before mating with

untreated males, as well as during pregnancy (3 weeks) and lactation (5 weeks).

Kidney was taken from the offspring at two stages (3rd

and 5th weeks) ages for the

current study. The exposure of rats to different doses of uranyl acetate caused a

significant (p<0.05) decrease b.w. of the offspring at 3rd

week age, while b.w of the

offspring revealed no significant at the 5th

week age, and also decreased the numbers

of the offspring . The histopathological changes in the kidney of the rat offspring

were revealed variety to the lesions like widening to the proximal convoluted tubule,

distraction to the tubular epithelium, and atrophy to the glomerulus, also widening to

the urinary space, and narrowing of the tubular lumen. Also it was showed hyaline

cast inside the lumen of urinary tubule, was shown with a necrosis of the tubular

epithelium, and vacuolation of the cytoplasm.

Introduction: Soluble uranium and all nano-particles can

cross the placenta, and these are particularly

toxic to the rapidly developing embryo or fetus

[1]. Very high doses of U in drinking water can

affect the development of the fetus in the

laboratory animals [2]. As well, U affects on

postnatal development and behavior of the

offspring [3].

The half-life of U in the kidneys of human has

been estimated to be 1 to 6 days for 99% of the

distributed dose and 1,500 days for the

remainder [4]. In rats, 95% of the dose

distributed to the kidneys is excreted in the urine

within one week [5,6,7]. The half-life of U

compounds in the kidneys has been reported to

be 2 to 17 days in rats [8,9]. In the kidneys, U is

released from bicarbonate and is free to combine

to form complexes with phosphate ligands and

proteins in the tubular wall to cause damage

[10]. Although the regenerated epithelium has

histological differences from its normal state, it

is often difficult to detect histological signs of

kidney damage a month after exposure because

all remaining functional damage is subtle. An

alternative mechanism through which U exerts

its renal toxicity has been suggested by the

results of a study conducted with rabbit kidney

cells in vitro. In some studies, uranyl nitrate

hexahydrate inhibited both sodium transport-

dependent and independent ATP utilization and

mitochondrial oxidative phosphorylation in the

renal proximal tubule [11, 10].

Many animal studies have shown that inhalation,

oral or dermal exposure to U results in kidney

damage. The damage was histological

manifested as glomerular and tubular wall

degeneration. Ultra structural analysis showed

damage to the endothelial cells in the

glomerulus, such as loss of cell processes, and

reduction in the density of the endothelial

fenestrate [12, 13, 14].

Leggett,(1989)(15)and Domingo, (1995) [16]

have been shown that uranyl ion inhibits Na+

transport-dependent and Na+ transport-

dependent ATP utilization, as well as

mitochondrial oxidative phosphorelation in the

renal proximal tubules, at doses not high enough

to destroy a critical mass of kidney cells. The

effect of the uranyl ion appears to be reversible,

as some of the cells which are replaced.

Materials and Methods: Uranyl acetate:

Dissolve powder of uranyl acetate in D.W,

and administrated with 50, 75,100 mg/kg/b.w

[17].

Animals and housing condition: Thirty (10 males and 20 females) sexually

mature laboratory breed males and females

Sprague- Dawley Albino rats (Rattus

norvegicus) of an average body weight of

230±3.565 gm and 12-15 weeks old were

obtained from animal house of the national

center for censorship and curative researches in

Baghdad.

Animals were kept in the Department of

Biology, College of Science, University of

Sulaimania; under the laboratory conditions (12

h light: 12h dark photoperiod), with controlled

room temperature 25-28oc, good ventilation and

were feed normal rodent pellets and tap water ad

Libitum.

Experimental Design:

The rats were randomly divided into four

groups. Each group were kept into box cage

measuring 40×25×15 cm. Female rats were

mated with males (2:1) until copulation was

detected. Finding of sperm indicated copulation

and the day of detection were designated as Day

0 of gestation. Then the adult fertile females

were treated with uranyl acetate dihydrate

(UAD) by gavages 10 days w before mating

with untreated males, as well as during

pregnancy and lactation [18]. The first group

served as a control and only receive drinking

water .However three concentration of uranyl

acetate dissolved in water were administrated

gavages to three other groups of females

(50,75,100 mg/ kg/ B.W/ day)

Dissection and Removal of Organ:

The offspring at the two stage of age (3rd

and

5th

weeks) were killed by cervical dislocation ,

then each group were scarified directly kidney

was taken from the offspring at the two stage of

age (3rd

and 5th

weeks).

Histological preparations Histological examination was done by fixing

the organs of the mice in 10% formalin saline,

processed and embedded in paraffin wax. Tissue

blocks were sectioned 5 µm thick and stained

with Haematoxylin and Eosin. [19].

Result: Body weight:

In this study, the b.w. of the offspring at 3rd

week age were significantly decreased compare

with the control , as in (Table 1). While, the b.w.

of the offspring at 5th week age were no

significant different, compare with the control,

as in (Table 1).

Histological changes:

A normal histological structure of the kidney

showed a normal structure of glomerulus with a

normal bowman's capsules and bowman's space,

also showed a normal structure of convoluted

tubules (Fig.1). Section of kidney of offspring at

5th and 3th week (G2) showed widening to the

renal tubule, atrophy to the renal corpuscle and

vaculation to the cells of the renal tubule (Fig. 2)

Discussion: In this study, significant weight loss was

noticed in the b.w. of the offspring at 3rd

week

age compared with the control, These results

are consistent with [20] the main reason of this

loss in b.w at this age is totally depending on

mother's milk which contain U.A from exposed

females [21], before that passes across the

placenta during the pregnancy [22]. While, the

b.w. of the offspring at 5th week age were not

significantly different compare with the control,

as in (Table1; Fig.1), because they were feeding

normal rodent pellets in addition of mother's

milk. Therefore, the immune system was in

progress at this stage [23]. The kidney is a

complex organ consisting of well-defined

components that function in a highly

coordinated fashion, when a segment of the

functional unit of the kidney is altered, acute

renal failure (ARF) may occur Tubules are the

most frequently damaged area in ARF [24]

vaculation of the cytoplasm and necrosis to the

renal tubular epithelium, and widening and

dilatation to the renal tubules, also vaculation to

congestion in the renal blood vessels, slid

widening to the urinary space, and atrophy to the

renal corpuscle and necrosis and vaculation to

the renal tubular epithelium, and atrophy to the

renal corpusels that’s agreed with [25] these

lesion translate from mothers to their offspring

cross placenta and during lactating as discussed

later. Section of kidney of offspring at 5th week

(G2) showed necrosis the proximal convoluted

tubular epithelium and focal distraction to the

renal parenchyma, also widening to the renal

tubule, and atrophy to the renal corpuscle and

vaculation to the cells of the renal tubule.

week (G2)

showed necrosis and distraction to the renal

tubules epithelium focal infiltration of the

inflammatory cells, and vaculation to the cells of

the renal tubule as shown in also, focal

infiltration of the inflammatory cell. Once in the

blood, U is distributed to the organs of the body.

Uranium in body fluids generally exists as the

uranyl ion (UO2)2+ complexes with anions such

as citrate and bicarbonate. Approximately 67%

of U in the blood is filtered in the kidneys and

leaves the body in urine within 24 hours; the

remainder distributes to tissues [20]. The

histopathology damage to the proximal tubules

in G4 and G3 is manifest as cytoplasmic

vacuolation, interstitial scarring, and destruction

of the basal lamina [26]. Histopathology

examinations showed that the UA causes sever

damage in the kidney of rats offspring, similar

results were recorded by [27, 16] who were

attributed that the U caused renal injury in the

experimental animal mostly the damaging of the

proximal convoluted tubules, however, it has

also been found to extend into other regions of

the kidneys, including the corticomedullary

region and the glomerulus [27, 28]. Also atrophy

to the glomerulus and widening to the urinary

space were observed in this study, Sclerosis of

glomerular capsules, reticulum sclerosis of

tubular basement membranes and interstitial

scarring, Dilation, apical displacement, and

vesiculation of tubular nuclei, glomerular

adhesions, focal tubular degranulation and

sloughing the cells into the tubular lumen, these

results were in agreement with previous results

of Gilman et al., (1998)[26].While, there is no

evidence for glomerular injury [29]. A

mechanism involving bicarbonate uptake in the

kidneys and subsequent precipitation of U in the

tubule was proposed for U-induced renal

toxicity [2]. Also, the results were revealed

focally severe necrosis of small lining epithelial

cells of some renal tubules as indicated by

sloughing of cells into the tubular lumens, loss

of nuclei and vaculation of the cytoplasm, were

same as [30] In general, histopathological

sections in G2(treated with 50 mg/kg/b.w)

reveals necrosis the proximal convoluted tubular

epithelium and focal distraction to the renal

parenchyma, also widening to the renal tubule,

atrophy to the renal corpuscle and vaculation to

the cells of the renal tubule. Mechanisms for

decreased glomerular filtration appear to involve

multiple factors including changes in renal

plasma flow, glomerular capillary hydrostatic

pressure, tubular hydraulic pressure, and

glomerular ultrafiltration coefficient. Tubular

impairments include glucosuria, amino aciduria,

enzymuria, and osmotic diuresis. Prominent

features of tubular injury are initial necrosis of

the terminal segments of the proximal tubule

with subsequent involvement of the distal tubule

[30]. Although specific studies on the

nephrotoxic effects of U in children have not

been conducted, it is reasonable to assume that

children would be at increased risk for adverse

effects from exposure compared with adults.

Children consume more water and food per

kilogram of body weight than do adults [31].

Thus, children will ingest proportionately

greater quantities of any contaminants that are

present in the water or food that they consume.

For example, the 3-year-old girl in this case

series who manifested elevated urinary excretion

of beta-2-microglobulin was reported to derive a

major portion of her nutritional intake from

infant formula that was prepared by mixing

powdered formula with contaminated well water

[32]. Alterations causing thickening of the

glomerular basement membrane of the kidney,

which results from the storage of uranium in the

kidney can be prolonged and severe enough to

cause permanent damage [33]. Sclerosis of

tubular basement membranes and interstitial

scarring, Dilation, apical displacement, and

vesiculation of tubular nuclei, glomerular

adhesions, focal tubular degranulation and

sloughing the cells into the tubular lumen, these

results were in agreement with previous
