RINGKASAN
Tanaman kelor (Moringa oleifera) merupakan bahan makanan lokal yang
memiliki potensi untuk dikembangkan dalam kuliner ibu menyusui, karena
mengandung senyawa fitosterol yang berfungsi meningkatkan dan memperlancar
produksi ASI (efek laktagogum). Secara teoritis, senyawa-senyawa yang mempunyai
efek laktagogum diantaranya adalah sterol. Sterol merupakan senyawa golongan
steroid (Nurmalasari, 2008).
Penelitian ini bertujuan untuk: (1) Mengukur kadar senyawa stigmasterol β-sitosterol dan kampesterol daun kelor, sehingga dapat dijadikan dasar untuk penanganan dan aplikasinya lebih lanjut pada produk pangan, khususnya pangan bagi ibu menyusui, (2) mencari dosis tepung daun kelor yang tepat dalam meningkatkan produksi air susu tikus putih galur wistar, sebagai langkah awal dalam mengembangkan model makanan ibu menyusui.
Penelitian ini dirancang dalam 2 tahap yang saling terkait. Diawali dengan tahap studi potensi daun kelor yang tumbuh di daerah Malang, selanjutnya dilakukan studi tentang potensi daun kelor sebagai bahan makanan yang dapat meningkatkan produksi air susu ibu (ASI), yang merupakan langkah awal dalam pengembangan model kuliner bagi ibu menyusui yang menggunakan daun kelor.
Hasil survey awal mengenai keberadaan dan pemanfaatan daun kelor ditinjau secara sosiobudaya yang dikumpulkan sebagai bahan untuk perancangan produk bagi ibu hamil dan menyusui di beberapa tempat, yaitu di Batu, Tumpang, Dampit, Junrejo dan Karangploso, diperoleh beberapa hasil berikut ini: (a) Daun kelor lebih banyak dimanfaatkan untuk memandikan jenazah, meluruhkan “ajimat”. Hal tersebut berdampak pada “tabu” untuk memanfaatkan daun kelor. Hal ini diduga karena di Indonesia, khususnya di Malang kurang informasi tentang khasiat daun kelor sebagai pelancar ASI, sulitnya buku referensi lokal yang membahas secara khusus mengenai tanaman kelor dan kegunaannya, tanaman Kelor sangat jarang dibahas dalam forum-forum ilmiah maupun media masa, terbatasnya model kuliner yang memanfaatkan daun kelor. Selain itu, faktor sosiobudaya sangat mempengaruhi konsumsi daun Kelor; (b) Daun kelor dimanfaatkan sebagai pakan ternak, sehingga ada pandangan inferior terhadap bahan makanan tersebut; (c) Hanya sedikit masyarakat yang memanfaatkan daun kelor sebagai sayuran. Kalaupun dimanfaatkan hanya diolah sebagai sayur bening saja dan (d) Masyarakat lebih banyak memanfaatkan buahnya sebagai sayuran.
Sebelum dilakukan uji kadar steroid pada tepung daun kelor, dilakukan
perlakuan pendahuluan awal pembuatan tepung terlebih dahulu. Perlakuan tersebut
berupa perlakuan blansing yang berbeda, yang meliputi rebus, kukus dan rebus+soda
kue. Hal ini dilakukan karena dalam pembuatan tepung akan terjadi perubahan sifat
organoleptil, kadar gizi yang tidak dikehendaki.
Berdasarkan hasil uji secara kualitatif tepung daun kelor terbentuk warna biru
ungu yang menandakan adanya kandungan senyawa steroid. Hasil tersebut diperkuat
dengan uji kuantitatif dengan menggunakan KCKT, menggunakan fase gerak methanol
: air (98 : 2) dengan kondisi elusi isokratik. Hasil yang diperoleh adalah: tepung daun
kelor dengan perlakuan awal blansing rebus memiliki kadar kampesterol 277,51 ppm,
stigmasterol 171,52 ppm. Tepung daun kelor dengan perlakuan awal blansing kukus
mengandung kampesterol 348,05 ppm, stigmasterol 2410 ppm dan β-sitosterol 3321,17
ppm. Sedangkan tepung daun kelor dengan perlakuan awal rebus+soda kue
mengandung kampesterol 278,81 ppm, stigmasterol 2113,43 ppm dan β-sitosterol
1635,95 ppm.
Hasil uji efek laktagogum pada tikus putih galur Wistar, diperoleh data sebagai
berikut: induk tikus yang diberi makanan tambahan tepung daun kelor dengan
perlakuan awal mencelur kukus melahirkan anak dengan rerata berat badan yaitu 5,6 g
pada pemberian dosis sonde seberat 42 mg/g bb induk tikus, 6,056 g pada pemberian
dosis sonde seberat 84 mg/g bb induk tikus dan 6,5 g pada pemberian dosis sonde
seberat 168 mg/g bb induk tikus. Sedangkan rerata peningkatan berat badan anak
tikus setelah umur 14 hari tertinggi pada anak tikus dari induk yang diberi sonde tepung
daun kelor dengan perlakuan awal mencelur kukus adalah 380,74% pada dosis 42;
500,9% pada dosis 84 dan 871,9% pada dosis 164 mg.
Simpulan dari hasil penelitian ini adalah : Daun kelor merupakan bahan makanan
yang dapat meningkatkan produksi air susu ibu. Hasil penelitian menunjukan
pemberian tepung kelor dapat meningkatkan produksi air susu induk tikus secara
signifikan. Pemberian dosis mulai 42 mg/kg bb secara signifikan dapat membuat
sekresi air susu tikus ptuih meningkat dan berat badan anak tikus meningkat seiring
dengan meningkatknya dosis yang diberikan.
SUMMARY
Plant Moringa (Moringa oleifera) is a local grocery that has the potential to be
developed in the culinary nursing mothers, because they contain phytosterol compound
that works to increase and facilitate the production of milk (laktagogum effect).
Theoretically, compounds that have the effect of which is a sterol laktagogum. Sterols
are steroid compounds (Nurmalasari, 2008).
This study aimed to: (1) Measuring levels of compounds β-sitosterol and
stigmasterol kampesterol moringa leaves, so it can be used as a basis for further
handling and their application in food products, especially food for breastfeeding
mothers, (2) to find the dose right Moringa leaf powder in increasing milk production
wistar rat strain, as a first step in developing a model of breast-feeding mother's diet.
The study was designed in 2 stages are interlinked. Beginning with the study phase
potential of Moringa leaves that grow in the area of Malang, then performed a study on
the potential of Moringa leaves as a food ingredient that can increase milk production
(milk), which is the first step in the development of culinary model for breastfeeding
mothers who use Moringa leaves.
Initial survey results about the existence and use of Moringa leaves collected
socioculture be reviewed as a material for designing products for pregnant and lactating
women in some places, namely in the Stone, Overlapping, Dampit, Junrejo and
Karangploso, obtained some of the following results: (a) Leaf Moringa is more widely
used for bathing the corpse, shed the "ajimat". This is impacting on the "taboo" to take
advantage of Moringa leaves. This is presumably because in Indonesia, especially in
Malang is less information about the properties of Moringa leaves as facilitating
breastfeeding, difficulties in the local reference books that discuss the particulars of
moringa plants and their uses, plant Kelor very rarely discussed in scientific forums and
mass media, the limited model Moringa leaf culinary use. In addition, factors greatly
affect consumption socioculture Kelor leaf, (b) moringa leaves are used as animal feed,
so there is an inferior view of the food material, (c) Only a few people who use Moringa
leaves as a vegetable. Even if used only as a vegetable processing nodes only and (d)
People tend to use fruit as a vegetable.
Steroid levels before the test performed on moringa leaf powder, made the
initial pretreatment flour first. Treatment in the form of different blanching treatments,
which include boiled, steamed and boiled + baking soda. This is done because in the
manufacture of flour will be changes organoleptic properties, nutrient levels are not
desired.
Based on test results in a qualitative form of Moringa leaf powder blue purple color
indicates the content of steroid compounds. These results are reinforced by a
quantitative test by using HPLC, using a mobile phase of methanol: water (98: 2) with
isocratic elution conditions. The results obtained are: Moringa leaf powder with boiled
blanching pretreatment levels campesterol 277.51 ppm, 171.52 ppm stigmasterol.
Moringa leaf powder with steam blanching pretreatment kampesterol containing 348.05
ppm, 2410 ppm stigmasterol and β-sitosterol 3321.17 ppm. Whereas Moringa leaf
powder with pretreatment contain baking soda + boil kampesterol 278.81 ppm, 2113.43
ppm stigmasterol and β-sitosterol 1635.95 ppm.
The test results laktagogum effect on Wistar rats, obtained the following data: the
mother rats were given food supplements with moringa leaf powder pretreatment
blanching childbearing steamed with a mean body weight was 5.6 g at dosing sonde
weighing 42 mg / g bb parent mice, 6.056 g of the sonde dosing weighing 84 mg / g mm
and 6.5 g of the parent mice at doses sonde weighing 168 mg / g bb parent mice. While
the average increase in weight of children aged rats after 14 days of the highest in
young rats fed the parent sonde Moringa leaf powder with steamed mencelur
pretreatment was 380.74% at a dose of 42; 500.9% at doses of 84 and 871.9% in dose
of 164 mg.
The conclusions from this research are: Leaf Moringa is a food that can increase
milk production. The results showed administration of Moringa powder can increase the
milk production of the parent mice significantly. Starting dose 42 mg / kg can
significantly make the milk secretion increased white mice and pup's weight increases
with increasing dose.
