Spektrometri masa adalah suatu teknik untuk
menghasilkan ion-ion dalam fase gas dari
molekul atau atom dalam sampel, memisahkan
ion menurut rasio masa per muatan, dan
mengukur kelimpahan dari ion yang terbentuk
Spektrometer masa merupakan instrumen
yang memisahkan atom, molekul, dan fragmen
molekul yang terionisasi dalam fase gas
berdasar pada perbedaan rasio masa per
muatan

PRINSIP MS:
MENGHASILKAN ION
Ionisasi Elektron : membombardir sampel dengan elektron
M = molekul analit, e- = elektron dan M+ = molekul analit terionisasi
M+ = ion molekular, m/z terkait dengan bobot molekul (dalam banyak kasus z = 1)
M+ merupakan kation radikal yang terbentuk karena kehilangan 1 elektron, memiliki cukup energi untuk mengalami fragmentasi menjadi ion dengan m/z lebih kecil

MENGHASILKAN ION
Ionisasi elektron menggunakan beda potensial sebesar 70 V
untuk mengakselerasi elektron, bagaimana sistem ini dapat
membuat molekul organik terionisasi dan terfragmentasi?
(EK = qV = zeV, energi ikatan molekul umumnya berada pada
kisaran 200 – 600 kj/mol)

MATRIX ASSISTED LASER DESORPTION

MALDI is achieved in two steps. In the first step, the
compound to be analyzed is dissolved in a solvent
containing in solution small organic molecules, called
the matrix. The second step occurs under vacuum
conditions inside the source of the mass spectrometer.

Quadrupole mass analyzer
• Kecepatan ion pada arah x adalah fungsi dari posisi sepanjang y
• Untuk suatu besaran rasio tertentu antara DC ke RF pada frekuensi tertentu hanya ion denga m/z tertentu yang akan sampai ke detektor
• Jika m/z dan frekuensi tidak sesuai dengan kondisi yang diminta persamaan, ion akan berosilasi dengan jalur yang lebar yang menyebabkan ion bertubrukan dengan batang atau tertarik oleh vakum.
• Hanya nilai m/z tunggal yang dapat melewati quadrupole pada kondisi yang diberikan mass filter
• m/z range 1-1000 Da

Most FTICR mass spectrometers use superconducting
magnets, which provide a relatively stable calibration over a
long period of time.
Although some mass accuracy can be obtained without
internal calibrant, mass accuracy and resolution are inversely
proportional to m/z, and the best accurate mass
measurements require an internal calibrant.
Unlike the quadrupole ion trap, the FTICR mass
spectrometer is not operated as a scanning device.
