Throughout my career as a Hematopathologist, I have been passionate about volunteering for professional societies, in various areas of education, professional development and advocacy. In my clinical practice, I had the opportunity to serve as medical director for clinical flow cytometry laboratories, at academic institutions and reference laboratories, for more than a decade, and to oversee laboratory test development in a complex regulatory environment. At the same time, I had the privilege of overseeing or contributing to professional development activities by working with several major professional societies (ASCP, CAP, SH and ICCS). For the past several years, I volunteered as a member of the ICCS Education Committee and also chaired the e-learning subcommittee. More recently, I became a member of the Advocacy and Outreach Committees at ICCS and will be co-chairing the Advocacy session at this year’s ICCS annual meeting. In addition to my work with these committees, I am also an ad-hoc contributor to the Newsletter, Q&A and Video subcommittees of the ICCS. Over the years, my strong interest in understanding the complex regulatory landscape, as it pertains to the clinical laboratory practice, and desire in promoting our profession to ensure a robust current and future pathology workforce, have provided me with the necessary background for continuing in a similar role at ICCS. Therefore, I would be honored and delighted to serve as a Senior Councilor for Advocacy for the Society. Horatiu Olteanu - Statement of Interest

Clinical Practice, Interests, and Accomplishments 1. Clinical education and training of residents, fellows, practicing pathologists, and laboratory professionals: A key clinical interest and practice of mine also includes the clinical education and training of future physicians, pathologists, and laboratory professionals. I have made several key contributions that provide guidance and education on how to practice medicine for our future pathologists and laboratorians. These contributions include co-authorship on a comprehensive textbook on Lymph Node Pathology, serving as Program Director for the Hematopathology fellowship, and as member of chair of several clinical practice and education committees of national and international Pathology organizations, including the Association for Clinical Pathology (ASCP), College of American Pathologists (CAP), Society for Hematopathology (SH), and International Clinical Cytometry Society (ICCS). As chair of ASCP’s Pathologist Recertification Individualize Self-Assessment Examination (PRISE) committee, I have overseen and implemented a highly successful clinical education program that over the last 5 years experienced an average of 580 unique participants for each of the 25-30 annual subspecialty modules published by the committee. In addition to practicing pathologists, over 60% of US Pathology residency programs have subscribed to this program, making it a valuable training tool for residents and program directors. 2. Immunophenotypic characterization of hematologic malignancies for relevant diagnostic and prognostic markers: My personal and professional goals have been and will remain a striving for diagnostic excellence in Hematopathology, and a close involvement in, and creation of an environment of enhanced learning for medical professionals, including pathologists and oncologists. Since my initial appointment at the Medical College of Wisconsin, I was grateful for having the opportunity to support the departmental and institutional goals of patient care and education, particularly in providing a diagnostic service which supported the Division of Hematology and Oncology in providing high-quality patient care. Since I took over as medical director, the MCW Clinical Flow Cytometry laboratory experienced a tremendous growth in testing volume, sustaining an average 10% increase/year. Under my supervision, the laboratory transitioned from 4-color to 8-color flow cytometry testing, which is particularly useful in providing more detailed diagnostic information derived from limited specimens and uses less antibodies, thus contributing to a decrease in operating costs. Our laboratory was the first one in the

state of Wisconsin to offer 8-color flow cytometry testing. We were also following patients treated at a busy bone marrow transplant center, including minimal residual disease analysis for plasma cell myeloma, acute leukemia, and lymphoproliferative disorders, as the first laboratory in the state of Wisconsin to offer these tests. My clinical expertise in flow cytometry and Hematopathology has resulted in several key publications on guiding therapy in patients with T cell lymphoproliferative disorders (Olteanu H, Harrington AM, Hari P, Kroft SH. Research Interests and Accomplishments 1. Biomarker discovery in hematologic neoplasms: Our group was the first to publish on the clinical and pathologic impact of diagnostic and prognostic markers such as CD23 and CD200 in follicular lymphoma, plasma cell myeloma, and plasma cell dyscrasias. Currently, I am continuing these studies by focusing on post-transplant lymphoproliferative disorders (PTLDs) and the interplay of regulatory T cells (Tregs), CD200 and CD30 expression, and EBV status on clinical outcome. We are employing a combination of tissue microarray immunohistochemistry, in situ hybridization, imaging studies and B-cell receptor gene rearrangement RT-PCR to address the utility of new biomarkers for prognostic and therapeutic purposes. Am J Clin Pathol, 20;148(6):485-493, 2017. 2. Gammaherpesvirus-induced lymphomagenesis in immunocompetent and immune-deficient individuals: I am collaborating with Dr. Vera Tarakanova from the Medical College of Wisconsin on a project which poses a clinically relevant interdisciplinary conundrum at the crossroads of molecularly relevant host factors (IRF-1 expression) and gammaherpervirus-driven lymphomagenesis. Prior to our studies, the role of IRF-1 in the regulation of gammaherpesvirus replication, chronic infection, and pathogenesis was poorly understood. Our group was the first to demonstrate that gammaherpesvirus infection synergizes with IRF-1 deficiency to induce B cell lymphomas. This innovative project is based on our observation that IRF-1 suppresses gammaherpesvirus-driven expansion of germinal center B cells; this is the first time IRF-1 has been implicated in virus-driven B cell differentiation and has resulted in intramural and extramural funding. Successful completion of the proposed studies will establish a novel and tractable animal model of gammaherpesvirus lymphomagenesis. The ultimate, long-term goal of our research is to determine the extent to which IRF-1-regulated gene expression suppresses gammaherpesvirus-induced cancer in humans. J Exp Med 2018 (submitted for review) 3. Immunophenotypic characterization of hematologic malignancies for relevant diagnostic and prognostic markers: Our group’s clinical research is focused on the application of specialized ancillary techniques, in particular flow cytometry, to the clinical and pathologic characterization of hematolymphoid malignancies. We were the first to publish on the temporal immunophenotypic changes of neoplastic cells in plasma cell myeloma and T-cell large granular lymphocytic leukemia, and to provide a detailed characterization of normal and aberrant NK cells in the post-transplant setting or in neoplastic conditions: Spears, M.D., Harrington, A.M., Kroft, S.H., Olteanu, H.: Immunophenotypic stability of T-cell large granular lymphocytic leukemia by flow cytometry.
