Kode genetik• Informasi dari DNA dikodekan ke dalam suatu
urutan tiga nukleotida (triplet nukleotida) pada
mRNA, yang masing-masing merupakan “kata
sandi” untuk satu asam amino yang spesifik.
• Tiap triplet pengkode itu disebut kodon.
• Ada 4 macam basa nukleotida pada mRNA
Adenine (A), Uracil (U), Guanine (G) and
Cytosine (C)
• Ada 20 asam amino di alam
jika tidak dikombinasi 4 basa nukleotida 4
AA
Jika dikombinasi menggunakan 2 nukleotida
42 = 16 AA
Jika dikombinasi menggunakan 3 basa
nukleotida (triplet) 43 = 64
cukup untuk dikode menjadi 20 AA
• Masing-masing asam
amino mempunyai
kodon yang spesifik.
• Dengan demikian
jumlah kode genetik
ini lebih banyak dari
jumlah asam amino
yang ada.
• Ternyata diketahui ada beberapa kodon yang mengkode
asam amino yang sama (genetic code redudancy atau
degeneracy).
• 61 kodon merupakan kodon sense yang mengkodekan
asam-asam amino. Satu diantaranya merupakan start
kodon yaitu kodon AUG (metionin) yang akan
menempatkan diri pada permulaan (ujung asam amino)
pada semua rantai polipeptida.
• 3 kodon tidak mengkode asam amino apapun karena
ketiganya merupakan kodon untuk mengakhiri proses
translasi, yaitu :
TAA (UAA pada mRNA) , TAG (UAG pada mRNA),
TGA (UGA pada mRNA). Disebut kodon nonsense atau
stop kodon.
• Dalam proses translasi dibutuhkan sekuen
triplet tRNA yang mencocokan mRNA untuk
sintesis .
• Satu-satunya cara untuk mencocokan
nukleotida tersebut adalah melalui base pair
yang akan saling melengkapi, triplet tRNA yang
mengkode dan melekat pada asam amino
spesifik disebut antikodon
• Berpasangannya kodon dan antikodon terjadi
di ribosom dimana asam amino yang
berlekatan dengan tRNA dapat digabungkan
oleh ikatan peptida membentuk polipeptida

Because codon-anticodon interactions can allow "wobble" pairing at the third position of a codon (the first position of an anticodon), it is possible for a single tRNA to recognize two or three codons. The allowable base pairing interactions are shown in the next slide and are listed above. This is accomplished because G can pair not only with C but also with U, and because of the modified base inosine. Inosine is derived from adenosine by deamination.
"Wobble" during reading of the mRNA allows some tRNAs to read multiple codons that differ only in the 3rd base.
